چکیده

It is well established that quantitative trait loci (QTL) underlying economically important traits can be mapped in livestock using existing marker maps either in experimental crosses or outbred populations. The resolution that can be achieved using conventional designs, however, is usually insufficient for positional cloning of the corresponding genes. We briefly describe approaches to fine-map QTL, with particular emphasis on an identity-by-descent method that was successfully applied to locate a QTL influencing milk yield and composition in cattle within a 5 cM chromosome segment. Methodological options to go from a fine-mapped QTL to the identification of the actual genes and mutations are envisaged.

A Successful Marriage: Making the Transition from Quantitative Trait Locus Mapping to Marker-Assisted Selection (Also published on-line in AgBiotechNet at URL http://agbio.cabweb.org)

The change from a Mendelian to a genomic breeding paradigm poses three challenges: to map the components of animal breeding onto a genomic conceptual frame work; to show how the genetic parameters comprising a genomic analysis can be obtained from actual farm data; and to show how genomic parameters can guide a breeding program. We are currently heavily engaged in the second challenge, but shou...

PGMapper: a web-based tool linking phenotype to genes

SUMMARY With the availability of whole genome sequence in many species, linkage analysis, positional cloning and microarray are gradually becoming powerful tools for investigating the links between phenotype and genotype or genes. However, in these methods, causative genes underlying a quantitative trait locus, or a disease, are usually located within a large genomic region or a large set of ge...

Linkage analysis of microsatellite markers on chromosome 5 in an F2 population of Japanese quail to identify quantitative trait loci affecting carcass traits

An F2 Japanese quail population was developed by crossing two strains (wild and white) to map quantitative trait loci (QTL) for performance and carcass traits. A total of 472 F2 birds were reared and slaughtered at 42 days of age. Performance and carcass traits were measured on all of the F2 individuals. Parental (P0), F1 and F2 individuals were genotyped with 3 microsatellites from quail chrom...

Creating Porcine Biomedical Models Through Recombineering

Recent advances in genomics provide genetic information from humans and other mammals (mouse, rat, dog and primates) traditionally used as models as well as new candidates (pigs and cattle). In addition, linked enabling technologies, such as transgenesis and animal cloning, provide innovative ways to design and perform experiments to dissect complex biological systems. Exploitation of genomic i...

Microsatellite mapping of quantitative trait loci affecting carcass traits on chromosome 1 in half-sib families of Japanese quail (Coturnix japonica)

The objective of this study was to identify the quantitative trait loci (QTL) affecting carcass traits on chromosome 1 in Japanese quail. The populations comprised of 422 progeny in 9 half-sib families. Phenotypic data on carcass weight, carcass parts, and the internal organs were collected on 422 progeny. Nine half-sib families were genotyped for 8 microsatellite markers covering chromosomes 1...
